摘要:【中文】一种丁香酚完全抗原的合成方法,本发明涉及化学物合成技术领域;丁香酚羧基化半抗原的制备;称取丁香酚羧基化半抗原,溶于1mL PBS缓冲液中;称取12mg载体蛋白溶于1mL PBS缓冲液中,得到载体蛋白溶液,然后将半抗原溶液逐滴加入到载体蛋白溶液,得到半抗原和载体蛋白混合溶液;将5mg EDC溶于0.5mL PBS缓冲液中,得到EDC溶液,然后将EDC溶液边搅拌边逐滴加入上一步得到的半抗原和载体蛋白混合溶液中,继续冰浴15min,室温下振荡反应1h,反应结束后,用PBS透析;对丁香酚完全抗原的鉴定。制备的丁香酚完全抗原弥补了丁香酚免疫学检测方法技术领域的空白,为丁香酚酶联免疫检测方法的进一步发展奠定了基础。 【EN】The invention discloses a synthetic method of a eugenol complete antigen, and relates to the technical field of chemical synthesis; preparing eugenol carboxylation hapten; weighing eugenol carboxylated hapten, and dissolving the eugenol carboxylated hapten in 1mL of PBS buffer solution; weighing 12mg of carrier protein, dissolving the carrier protein in 1mL of PBS buffer solution to obtain a carrier protein solution, and then dropwise adding the hapten solution into the carrier protein solution to obtain a mixed solution of the hapten and the carrier protein; dissolving 5mg of EDC in 0.5mL of PBS buffer solution to obtain EDC solution, then dropwise adding the EDC solution into the mixed solution of the hapten and the carrier protein obtained in the previous step while stirring, continuously carrying out ice bath for 15min, carrying out oscillation reaction at room temperature for 1h, and dialyzing with PBS after the reaction is finished; and (3) identifying a eugenol complete antigen. The prepared eugenol complete antigen makes up the blank of the technical field of eugenol immunological detection methods, and lays a foundation for the further development of eugenol enzyme-linked immunosorbent assay methods.
