摘要:【中文】本发明属于马铃薯病毒领域,尤其是一种马铃薯Y病毒检测方法,其包括以下步骤:S1:取0.1g带有症状的新鲜烟叶,放入提前用液氮预冷的灭菌研钵中,加入液氮充分研磨,直至叶片呈白色粉末状,之后迅速将粉末转移至液氮预冷的2ml离心管内;S2:在样品粉末潮解前迅速加入Trizol,充分混匀,再加入氯仿,颠倒混匀,冰上静置;S3:离心机提前开启调至4℃,离心15min,本发明丰富了人们对WTMV的了解,并获得了其全基因组核酸序列,有利于人们研究WTMV的遗传信息及种群遗传分化,这对于及早了解WTMV的分布、流行及防控有一定的帮助,为以后农业的发展奠定了基础。 【EN】The invention belongs to the field of potato viruses, and particularly relates to a potato Y virus detection method, which comprises the following steps: s1: taking 0.1g of symptomatic fresh tobacco leaves, putting the symptomatic fresh tobacco leaves into a sterilization mortar pre-cooled by liquid nitrogen in advance, adding the liquid nitrogen to fully grind until the leaves are white powder, and then quickly transferring the powder into a 2ml centrifugal tube pre-cooled by the liquid nitrogen; s2: rapidly adding Trizol before deliquescence of the sample powder, fully and uniformly mixing, adding chloroform, reversely and uniformly mixing, and standing on ice; s3: the centrifuge is started to adjust to 4 ℃ in advance, and the centrifuge is centrifuged for 15min, so that the WTMV knowledge is enriched, the whole genome nucleic acid sequence is obtained, the genetic information and population genetic differentiation of the WTMV can be conveniently researched by people, the WTMV distribution, popularity and prevention and control can be known as soon as possible, and a foundation is laid for future agricultural development.
